Auxotrophic Strains

 

Auxotrophic Strains of P. tricornutum would reduce the use of traditional antibiotic selection, enable both positive and negative selection, and are essential for biocontainment of engineered P. tricornutum. Currently, known auxotrophic strains are:

 

  • -HIS

  • -URA

  • -TRP

A recent preprint by Slattery et al. presents a Cas9-generated method of generating a -URA strain of P. tricornutum.
